Proinflammatory cytokine tumor necrosis factorα (TNF-α) contributes to a variety of inflammatory responses and programmed cell death. Notch receptor and its ligand participate in cell fate decisions during vertebrate development and are associated with several human disorders, including a T-cell lymphoma. TNF-α, notch and its ligand delta are all membrane-bond molecules, which are cleaved by proteases to release mature proteins or functional receptor. ADAM10, a metalloprotease-disintegrin in the family of mammalian ADAM (for a disintegrin and metalloprotease), was recently identified to cleave TNF-α, notch and its ligand delta. The genes encoding human, mouse, and bovine ADAM10 were recently cloned and designated ADAM10, kuzbanian (KUZ), and MADM, respectively. ADAM10 mRNA is expressed in a variety of human and bovine tissues.

100 µg (0.5 mg/ml) immunoaffinity purified rabbit anti-ADAM10 polyclonal antibody in phosphate buffered saline (PBS), pH 7.2, containing 30% glycerol, 1% BSA, 0.02% thimerosal.
This antibody has been succesfully tested for use in following methods:
Western blotting (1-4 µg/ml) and immunofluorescence (2-10 µg/ml). An 85 kDa band corresponding to the precursor form of ADAM10 can be detected. Jurkat and K562 cell lysates can be used as positive controls. However, the optimal conditions should be determined individually.

The antibody solution should be gently mixed before use.
This antibody needs to be stored at + 4°C in a fridge short term in a concentrated dilution. Freeze thaw will destroy a percentage in every cycle and should be avoided.
